COMMENT The proximal of 1.6 kb of the 5'-flanking region of SpMTA
metallothionein (MT) gene together with the 1.12 kb first intron
are sufficient for its correct cell-type specific expression in
transgenic embryos [2,3]. The heavy-methal ion-dependent activity of
promoter is sustained through the cooperative interaction of
a series MRE sites [1]. The regulatory elements in intron enhance as
well as restrict expression to the single set of embryonic cell
lineages [2]. The restricted spatial expression is largely eliminated
by the deletion of the interior 405-bp region in the intron.
Within this region is 295-bp, genomically repetitive,
transposon-like segment [2,3], containing P3A site and four
apparent P5 sites, two of which are embeded in
25-bp inverted repeats at its termini. The P3A and terminal P5 sites
were shown [3] through competition to bind with relatively high
affinities the same nuclear factors bound by their counterpart
sites in the CyIIIa actin promoter
