REFERENCE 1 (bases 1 to 348) AUTHORS Mao, C. A., Gan, L., Klein, W. H. TITLE Multiple Otx binding sites required for expression of the Strongylocentrotus purpuratus Spec2a gene. JOURNAL Dev. Biol. 165 (1): 229-242 (1994) MEDLINE 94374572 REMARK GeNet staff at the IHC&DB created this entry from the original journal article. This sequence comes from Fig. 1B COMMENTS The RSR enhancer is a 188-bp SstI-XhoI between base pairs -631 and -443 (154-342 of this entry) in the Spec2a control region. The RSR enhancer appears tp be a key regulatory region controlling Spec2a gene transcription during embryogenesis. Three DNA elements with the consensus sequence TAATCC/T are positive cis-regulatory elements essential for full reporter gene expression in sea urchin embryos. Because the sea urchin orthodenticle-related protein SpOtx binds these TAATCC/T elements with high affinity, they have been designated Otx sites. These sites, and most likely a fourth just upstream of the RSR enhancer, appear to act additively to confer positive activity to the Spec2a control region [1].
